SX20 Stopped-Flow Spectrometer Accessories - Fluorescence Polarization

SX.20 Stopped-Flow Spectrometer

FP.1 Fluorescence Polarization/Anisotropy

FP.1 extends the measurement possibilities of the SX range of stopped flow spectrometers by providing a convenient way to record steady state and kinetic fluorescence polarization and anisotropy data.

Excitation of a fluorophore with plane polarised light results in the preferential excitation of molecules with their absorption moments orientated parallel to the plane of polarization. Fluorescence polarization/anisotropy can provide information about changes in the mobility and environment of a fluorophore when it interacts with other molecules.

SX.20 FP.1 Fluorescence Polarization Accessory SX.20 Fluorescence Polarization data

The FP.1 fluorescence polarization/anisotropy accessory is an easy to fit, dual channel T-format fluorescence polarimeter with a movable calcite input polariser. G-factor determination is controlled from the software and both kinetics and spectra may be acquired in polarization, anisotropy, total emission and raw data modes, and with full post-acquisition conversion between data modes as required.

Note: option FP.1 includes both options DF.1 and DD.1.

Key Features of FP.1

  • T-format dual channel detection, greatly reducing noise contributions from the light source.
  • Calcite excitation polariser, enabling excitation from 220nm upwards.
  • DPUV sheet collection polarisers, enabling detection of polarised fluorescence from 300nm upwards.
  • Filter holders built into collection pieces, allowing reduction of scattered excitation light.
  • Filter holder built into excitation assembly, allowing highly versatile wavelength selection and rejection.
  • Mountings for end window or side window PMTs.
  • Easily mounts onto your SHU cell block.
  • Robust construction ensures consistent polariser alignment.

Software Features

  • Polarisation, Anisotropy or Total Emission data formats available.
  • Data may be collected in raw form, processed form, or both.
  • Data collection possible in kinetic, scanning and kinetic scanning modes.
  • Automatic PMT targeting and G-factor correction built into software.
  • Offset correction enables total removal of residual scatter contributions.
  • New transform functions specifically provided for processing of raw data and inter-conversion of polarization and anisotropy.
